TitleAffinity chromatography with ConBr lectin (Canavalia brasiliensis) for glycoprotein purification and characterization from COVID-19 patient serum
DescriptionSerum samples from COVID-19 patients (severe, mild, symptomatic non-COVID, and negative controls) were subjected to albumin depletion and affinity chromatography using ConBr lectin (Canavalia brasiliensis) coupled to sepharose matrix and a commercial ConA matrix (HiTrap ConA Sepharose 4B, Canavalia ensiformis), both specific for glucose/mannose. Eluted fractions were analyzed by nano-LC-MS/MS on an Orbitrap Velos system (Easy nLC1000, EASY-Spray PepMap C18 50cm x 75um, 300 nL/min). Data-dependent acquisition (DDA, top-20) with label-free quantification (LFQ) was performed. Database search used Sequest HT in Proteome Discoverer v3.1 against SwissProt Homo sapiens (TaxID:9606) and SARS-CoV-2 (UP000464024), with Carbamidomethyl (C) as fixed modification and Oxidation (M) as dynamic modification. Ethics approval: CEP CEUMA No. 209/2021-ESP/MA.
