TitlePROTEOGOLD: Generation of Reference MS/MS Datasets for Large Protein Using FTMS Simulation
DescriptionTop- and middle-down (TD/MD) mass spectrometry is widely used to characterize proteins and biotherapeutics. These workflows routinely produce highly complex, information-dense spectra—overlapping isotopic patterns, multiple charge-state envelopes, and crowded product-ion populations—that increasingly challenge existing data processing and interpretation tools. Robust evaluation of such tools, however, is hindered by the limited availability of “gold-standard” datasets that capture realistic experimental complexity across instruments, acquisition conditions, and analyte classes while providing known ground truth. Here we present simulated TD-MS/MS spectra for carbonic anhydrase II generated within the ProteoGold dataset initiative. By producing in-silico spectra under defined conditions, ProteoGold provides extensible reference datasets designed for benchmarking, education, and method development in TD/MD MS.
