TitleIsolation and proteomic analysis of intracellular vesicles from the potato late blight pathogen Phytophthora infestans
DescriptionPhytophthora infestans is a major pathogen of potato and tomato, a key determinant to the successful outcome of disease laying with the effector proteins secreted by the pathogen during infection; these collectively act to facilitate evasion of plant immunity, physical breaching of the plant tissue, and nutrient acquisition by the pathogen. These effector proteins can act within the host apoplast/extracellular space (apoplastic effectors) or are translocated into, and act within, the host cell (cytoplasmic effectors). There has been much interest in the fate and function of these proteins once secreted into the host plant, with the intracellular trafficking of these proteins enroute to secretion neglected. We combined differential centrifugation with cushioned bottom loaded gradient ultracentrifugation to isolate intracellular vesicles from in vitro grown P. infestans based on buoyancy. A transgenic P. infestans line expressing two fluorescently tagged effector proteins, PITG_04314-mCherry and PITG_01029-mCitrine, representing potential intracellular vesicle cargo enabled identification of the gradient fractions to which these proteins migrated. Mass spectrometry analysis of gradient fraction containing the majority of the two tagged effector proteins showed significant association with proteins associated with membranes and membrane trafficking. The method also identified the presence of known P. infestans extracellular vesicle markers (three tetraspanins and two MARVEL domain proteins), suggesting the capture of vesicles enroute to extracellular secretion.
