TitleMass Spectrometry-Based Peptidomics for Discovery and Profiling of Antimicrobial and Neuropeptide in Crustacean Hemolymph
DescriptionCrustacean hemolymph contains a diverse profile of bioactive peptides, including antimicrobial peptides (AMPs) and neuropeptides, which regulate essential immune and physiological processes. However, rapid coagulation and high-abundance proteins like hemocyanin complicate mass spectrometry-based peptidomics analysis. We systematically evaluated five extraction workflows to optimize endogenous peptide recovery from the hemolymph of the American lobster (Homarus americanus) and the blue crab (Callinectes sapidus). Among the tested approaches, Method 3, incorporating an anticoagulant buffer with protease inhibitors followed by 10 kDa molecular weight cutoff filtration, provided the highest peptide ID recovery. This workflow minimized clotting-related peptide loss and enzymatic degradation, yielding significantly greater numbers of AMPs in both species. Notably, omitting the reduction and alkylation protocol in Method 3 was superior for AMPs identification. For example, in C. sapidus, adding these steps (Method 4) resulted in a 40% decrease in AMPs IDs, likely due to increased sample handling and incubation times. Method 3 also maximized neuropeptide recovery in H. americanus, identifying mature orcokinins and myosuppressins. In C. sapidus, neuropeptide recovery varied across methods with Method 3 showing competitive performance. Additionally, we reported six novel putative C. sapidus neuropeptides, and identified 11 and 12 in silico predicted novel AMPs in C. sapidus and H. americanus, respectively, characterized by amino acid motifs. Collectively, this study demonstrated an integrated extraction strategy to enable more comprehensive profiling of crustacean hemolymph peptidomes and provided the first report of AMPs, alongside neuropeptides, in C. sapidus and H. americanus, expanding the repertoire of candidates for antimicrobial therapeutics and neuromodulation.
