TitleAbundance of the major extracellular protease Jep is stress-responsive in the mouse-adapted Staphylococcus aureus JSNZ - Cellular fraction
DescriptionThe opportunistic pathogen Staphylococcus aureus colonizes a broad spectrum of different hosts and niches. In recent years it was shown that mouse-adapted S. aureus strains are more suitable candidates to investigate S. aureus infections in the in vivo mouse model. One of the most studied mouse-adapted strain is JSNZ. Under control conditions at 37C, JSNZ secrets the highly abundant serine protease Jep (JSNZ extracellular protease), which is located on phi_LabRoddCC88_3. Due to the ability of S. aureus to colonize a broad variety of niches, we opted to investigate Jep expression under several physiological stresses such as different temperatures (37C, 42C and 32C) and limiting conditions (iron and oxygen limitation) on transcript and protein level. In silico predictions and Northern blot revealed a that jep is encoded monocistronic with an approximate transcript length of 900 bp. In the different stress conditions, jep transcript levels increases over time in a temperature-dependent manner. Contrastingly, secreted Jep increases in all conditions over time reaching a plateau in stationary phase. Mass spectrometry analysis proves that Jep is the major protein in JSNZ supernatant making up to 75% not only at 37C but also at 42C and under iron-limitation. The protein level of Jep correlates highly with other secreted proteases of JSNZ. Jep shows a phage-independent regulation. Analysis of co-regulated regulons to Jep and additional in silico analysis revealed SaeR and PurR as potential regulators of jep.
