| Title | The degradable kinome of DCAF1-targeting PROTAC |
| Description | Proteolysis targeting chimeras (PROTACs) have emerged as novel drug modalities, but the development of PROTACs currently relies on a limited number of E3 ligase ligands, primarily targeting CRBN and VHL. Validating novel E3 ligase ligands could expand the degradable target space. However, validation data on these understudied degrader systems are typically limited to a few highly degradable targets, such as BRD4. In the present study, we used our previously established workflow for E3 ligase ligand validation, employing promiscuous kinase PROTACs, to evaluate the potential of targeting DCAF1 for PROTAC development. Due to their similar architecture, we compared the degradable kinome of DCAF1 with an analogous set of CRBN-targeting PROTACs. Our study revealed DCAF1-dependent degradation of a diverse set of kinases, which were validated in orthogonal assays. We found a significant overlap of the degradable kinome with CRBN recruiting PROTACs, suggesting an alternative design strategy using available structurally diverse DCAF1 ligands. Furthermore, a comparative analysis of DCAF1 and CRBN PROTAC-induced ubiquitination patterns provided insight into substrate selectivity and isoform-selective degradation. The presented data will establish DCAF1-targeting PROTACs as a versatile design strategy for future degrader development. |
