| Title | Plasma Proteomic Characterization of Adolescent Idiopathic Scoliosis |
| Description | Adolescent idiopathic scoliosis (AIS) is the most common spinal deformity encountered in adolescents. Here we portray the plasma proteomic landscape of AIS samples. Enrichment analysis demonstrate that AIS-enriched proteins are significantly enriched in pathways including muscle weakness, disorder of hormone, whereas proteins enriched in healthy controls are mainly involved in pathways related to immune response. Alterations of tissue-enhanced proteins in plasma reveals AIS shows diverse impacts on organs, especially skeletal muscle and lymphoid. The WGCNA analysis indicates unbalanced lipid and glucose metabolism due to the insulin signaling activation could affect the AIS progression. Molecular subtyping classifies AIS patients into three subtypes (C1-C3) that connected with significantly different Cobb angle. C1 exhibit dysregulation in the reverse cholesterol transport and estrogen receptor binding; C3 is enriched in glucocorticoid biosynthetic process proteins, has the largest Cobb angle. Immune landscape characterization reveals AIS patients with larger Cobb angle have lower immune level, which are associated with higher glucocorticoid signal and Tregs levels. Additional, non-linear associations between Cobb and plasma proteome data reveals that the plasma proteome of 26 degrees and 51 degrees is dramatically differed across these two Cobb ranges. Finally, we construct a proteomics classifier that can distinguish AIS from healthy control (receiver operating characteristic–area under the curve, 0.94). This study not only generates a high-quality data resource that may benefit basic research but also provides additional biological insights underlying clinical features of AIS. |
