| Title | In vivo protein N-terminal sequence analysis of WAGO-1 and WAGO-3 in C. elegans |
| Description | A mass spectrometry-based label-free proteomics analysis was performed to study the N-terminal protein sequences of WAGO-1 and WAGO-3 in C. elegans. Argonaute-enriched proteins were digested by trypsin. Peptides were measured on an Orbitrap Astral mass spectrometer. Raw data files were processed by MaxQuant software package (version 2.6.2.0) using its integrated Andromeda search engine. Spectral data were searched against a target-decoy database consisting of the forward and reverse sequences of the WormPep release WS292 (28,587 entries), the UniProt E. coli K-12 reference proteome (release 2022_03; 4,460 entries), a list of N-terminally truncated WAGO-1/WAGO-3 proteins, WAGO-3 (P2G, A3E) mutant and a default list of common contaminants. False discovery rate (FDR) was set to 1% at both peptide and protein levels. |
