| Title | Ultra-deep chemoproteomic profiling of transglutaminase-mediated glutamine polyamination |
| Description | As key functional units in biology, proteins display activity, expression, localization, and structural features that are strongly influenced by their active sites. Despite the critical role of these sites, over 80% of human proteins remain classified as undruggable. The mapping of functional sites has therefore emerged as a key driver of first-in-class drug discovery. While chemical proteomics has revolutionized the study of drug–target interactions, research on glutamine active sites remains limited due to the low inherent reactivity of its side-chain amide under physiological conditions. In this study, we introduce an integrated chemical proteomics strategy using transglutaminase to systematically map glutamine active sites. |
