Unlike conventional Congo Red (CR), which only provides non-covalent staining and cannot use for protein enrichment, our CR-alkyne probe utilizes a boron-azo bioconjugation chemistry to form irreversible bonds with amyloid-associated proteins. This critical covalent linkage enables the specific enrichment of labeled proteins via the alkyne handle for LC-MS/MS analysis. The dataset directly contrasts with background proteins identified from control samples treated with native CR, which lacks the alkyne handle and covalent enrichment capability. This comparative design rigorously distinguishes true amyloid plaque constituents from non-specifically bound proteins, providing a high-confidence profile of the amyloid proteome and its interactors for understanding AD pathology. |
