TitleBN-SDS PAGE of Complex I from Phaeodactylum tricornutum
DescriptionA native complex I-enriched fraction isolated from Phaeodactylum tricornutum was separated by two-dimensional Blue-native / SDS Polyacrylamid Gelelectrophoresis (2D BN / SDS PAGE). After vizualization of proteins on the gel with Coomassie, protein spots were cut, cysteine residues were carbamidomethylated and an in-gel digestion was carried out with trypsin overnight. Peptides were extracted and used for HPLC-tandem MS using an online EvoSep (Evosep Biosystems) - timsTOF pro MS (Bruker) system. Data were analyzed with MaxQuant (Version 2.6.8.0) and further processed in Excel (Microsoft Office). The final processed dataset (which solely focusses on complex I subunits) was used to generate a reference map at www.gelmap.de.
