| Title | Biogenesis of the respiratory chain is controlled by de novo synthesis of cardiolipin in neonatal mouse hearts |
| Description | Postnatal maturation of mammalian heart requires a vast increase in respiratory enzymes. Because the mitochondria-specific lipid cardiolipin (CL) strongly interacts with the respiratory chain, we determined how the two steps of CL biogenesis, de novo synthesis and acyl chain remodeling, affect the maturation of cardiac mitochondria in mice. Cardiomyocyte-restricted deletion of CL synthase in late gestation did not affect CL at birth but blocked the steady increase in the tissue concentration of CL, which we observed during normal postnatal maturation. Deletion of CL synthase also prevented the postnatal rise in the density of cristae and in the intra-mitochondrial concentration of respiratory proteins. This inhibited cardiac development, precipitated heart failure, and caused death by the age of 2 weeks. In contrast, ablation of CL remodeling by cardiomyocyte-restricted deletion of Tafazzin, did not disrupt mitochondrial maturation or cardiac development although it had a similar effect on the CL concentration. Our data show that CL synthesis, but not CL remodeling, controls the expression of the respiratory chain by a mechanism independent of the CL concentration. |
