| Title | A method for profiling cell type-specific secretome: the airway microfold cell |
| Description | Protein secretion is a fundamental mechanism by which cells communicate with one another and shape their environment. Microfold (M) cells are rare, specialized cells in the mucosal epithelium which sample luminal antigens, channel them to the underlying mesenchyme, and regulate the mucosal immune system. Here we set out to uncover the secreted protein factors which govern the function of recently identified airway M cells by profiling the M cell secreted proteome (secretome). We generate a new mouse model which allows for the targeted profiling of the secretomes of cells of interest without the use of viral vectors. We then employ this new model to study the secretome of airway M cells in air-liquid interface (ALI) cultures. Our method demonstrates high specificity for M cells despite the complex mixture of cells in our cultures. Additionally, the method is characterized by remarkable sensitivity, capturing proteins from a rare cell that were not represented in prior scRNAseq airway atlases. Furthermore, since ALI cultures preserve epithelial polarity, we were able to distinguish secretion from either the luminal or basolateral cell surfaces. Together, these data present the first comprehensive, unbiased atlas of the airway M cell secretome, generating unique molecular insights into M cell function. We employ airway epithelial culture platforms as a proof of principle that can be extended to any organ system and make this mouse model available to the community to empower future studies of cell-to-cell communication, secreted protein function, and biomarker discovery. |
