TitleCD177 deficiency defines a stable subtype of human neutrophil granulocytes
DescriptionCD177 is a surface protein on human neutrophil granulocytes whose function is related, but not limited to, tissue infiltration. While a fraction of the population is completely CD177-deficient (CD177d), most individuals express CD177. Expression levels on cells are not uniform and in every individual a proportion of neutrophils is CD177d. Evidence is increasing that CD177 expressing (CD177e) neutrophils differ functionally from CD177d-cells and influence disease outcomes. Whether CD177e is a status ultimately reached by any neutrophil, also initially CD177d-cells, is unclear. This study characterized these two subtypes using LC-MS based label free quantitative proteomics. The proteomics data suggests distinct proteome composition of these two subtypes despite of having morphological similarity. Interestingly, CD177d-cells were found to present an N2-polarized phenotype based on protein expression, which are preferentially enriched in human head-and-neck cancers.
