| Description | Red blood cells (RBCs) play a vital role in adaptations to hypoxia at high altitude, however, metabolic alternations of RBCs under long-term hypoxia remain unknown. Method: Here, we employed the broad- and energy-targeted metabolism approaches to study the differences in the RBCs of lowlanders living in plain area and acclimated to plateau for more than 10 years, and adaptive plateau highlanders. Compared with plain individuals, the glycolytic flux remained stable in acclimated and adaped subjects, and purine metabolism was not correspondingly increased. Notably, the oxidative pentose phosphate pathway was reduced in the RBCs of acclimated and adapted sub-jects. The consistency loss was observed in the reduced glutathione, a key metabolite of the red blood cell redox system. The adaptive enhancement of glutamate amidation may indirectly contribute to the decrease in reduced glutathione. Further, reduced superoxide dismutase activity and enhanced catalase activity confirmed a distinct al-teration in the antioxidant enzyme system of RBCs under long-term hypoxic. Metab-olomics and clinical measurements support the view that RBCs undergo lower oxida-tive stress under hypoxia, offering new insights into the hypoxic adaptations. |
