| Title | Biological Characterization of Helicobacter pylori Outer Membrane Vesicles (OMVs) |
| Description | Helicobacter pylori is known to be a major pathogen causing gastric diseases via its direct colonization of the gastric mucosa. In this study, outer membrane vesicles released by H. pylori grown in planktonic and biofilm phenotypes at 2-6-10 days were collected and analyzed by LC-MSMS. The collected extracellular vesicles were lysed by adding a minimal amount of buffer containing urea 8M, tris 10 mM, dithiothreitol 50 mM, SDS and Triton X-100 and centrifuged at 13,000g for 15min to remove the insoluble fraction. The protein content was quantified through the Bradford assay and 30 microg of proteins were loaded onto a Nanosep 10-kDa-cutoff filter and trypsin digested according to our routine protocol. Each digested protein sample was analyzed in technical duplicate. Raw data were processed using PEAKS studio Xpro using the correct precursor only option. Spectra were matched against the UniProt SwissProt database restricted to H. pylori taxonomy, to which a list of common contaminants was appended. |
