TitleMembrane proteome profile of MSI-tumor organoids exposed to conditioned media of MSI and MSS-tumor organoids
DescriptionMSI-tumor organoids were previously stimulated with 200 ng/mL IFNγ for 24 hours. Then, organoids were dissociated into single cells using TrypLE express and resuspended in conditioned media. Cells were seeded in a 24-well plate and incubated for 24 hours at 37°C. After incubation, tumor cells were collected, washed in PBS and processed for membrane protein extraction using the Mem-PER™ Plus Membrane Protein Extraction Kit (Thermo Scientific, cat. no. 89842), following the manufacturer’s instructions. Membrane proteome was quantified using the Pierce™ BCA Protein Assay Kits (Thermo Scientific, cat. no. 23225). Then, the membrane protein fraction was digested for LCMSMS analysis.
