TitleProteins co-immunoprecipitated with the COPI complex
DescriptionWI-38 fibroblasts were lysed in RIPA buffer supplemented with protease/phosphatase inhibitor (Thermo Fisher Scientific), incubated on ice for 10 minutes, and centrifuged for 20 minutes at 13,200 rpm and 6 degrees Celsius. The supernatant was collected, and 1 mg of protein lysate was incubated with 10 ug of antibody (COPB1, COPE, COPZ1, all from Protein Tech, and IgG from Cell Signaling Technology) and 50 uL of DynabeadsTM protein G (Thermo Fisher Scientific 10004D). After overnight incubation at 4 degrees Celsius on a spinning rotor the beads were washed 5 times with RIPA buffer. Immunoprecipitated samples were processed for in-solution trypsin/LysC digestion and analyzed by Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) (Poochon Scientific). The resulting MS data was searched against the human protein sequence database using Proteome Discoverer 2.5 (Thermo Fisher Scientific).
