Title	Identification of active Papain-Like Cysteine Proteases in the third trifoliate leaves of Phaseolus vulgaris exposed to water deficit
Description	Two common bean genotypes (S156 and R123) with contrasting levels of resistance to environmental stresses, were exposed to a controlled terminal drought at mid-pod filling. The experiment was conducted in two identical FHC-2300 growth chambers (Hi-Point, Tawain). Climatic conditions were set at 300 µ m−2 s−1 for the Photosynthetic Photon Flux Density (PPFD), 14h of light per day for the photoperiod, 25℃ /22℃ for the day/night temperature and 70%/60% of relative humidity (HR). Mesocosms were arranged in a completely randomized design with their position weekly randomized. Starting at 42 days after sowing (DAS) and until pod maturity (100 DAS), for each genotype, half of the plants were randomly selected and subjected to a terminal drought. Terminal drought (Drought) was applied by limiting the water content at 19±1.4% of the plant extractable soil water content while the control plants (Control) were continuously watered at 73±0.5% of the plant extractable soil water content. DCG-04 probe, a biotinylated version of protease inhibitor E-64, was used to profile active papain-like cysteine protease (PLCPs) in the third trifoliate leaves of genotype S156 under control and drought conditions at 50 DAS (eight days after the start of drought treatment). Active PLCPs were enriched using an avidin purification method. Eluted proteins were separated by SDS-PAGE on 12% polyacrylamide Bis-Tris gels. Gels were stained with Coomassie blue G-250 and six slices corresponding to labelled proteins with a molecular mass between 25 and 40 kDa were cut out, digested by trypsin and analysed by LC-MS/MS.
