TitleUntargeted NoPro-clipping from untramsformed and Hoil-1L over-expressing Huh7 cell lines
DescriptionNoPro-clipping is a method for detecting and quantifying non-proteinaceous ubiquitination from cell lysates and mammalian tissue. The method utilising Ub-clippases to cleave ubiquitin but leave the C-terminal diglycine remnant attached to the protein and non-protein substrates. Following protein exclusion, small molecule diglycine species are selectively labelled with an LPXTGG-containing peptide via the enzyme sortase. While the method can be employed in a targeted workflow to quantify a pre-defined non-protein ubiquitin species, the method can also be employed in an untargeted approach. This submission includes the necessary controls and raw files to identify non-protienaceous ubiquitination present in Huh7 cells either untransformed or over-expressing the E3 ligase HOIL-1L fused to a di-ubiquitin (for increased activity).
