TitleProximity labelling of the post-Golgi AP complexes in Plasmodium falciparum
DescriptionAdaptor protein (AP) complexes are pivotal components of the post-Golgi trafficking and endocytosis process and are involved in the selection, package and sorting of cargo into vesicles to different destination compartments in eukaryotic cells. The main objective of this project was to investigate the proxiome of the post-Golgi adaptors AP-1, AP-3 and AP-4 in the human malaria parasite Plasmodium falciparum, using the DiQ-BioID approach. To this end, malaria parasites were genetically modified via selection linked integration (SLI) to express endogenously 2xFKBR-GFP-2xFKBP-tagged versions of the μ subunits of AP-1, AP-3 and AP-4 complexes, followed by transfections with plasmids to episomally express miniTurbo-FRB-mCherry (N-terminal) and mCherry-FRB-miniTurbo (C-terminal). The conditional recruitment of the miniTurbo to AP foci (trans-Golgi localisation) was induced by addition of rapalog. Consequently, a mass spectrometry analysis of biotinylated proteins resulted in a list of high-confidence hits for each adaptor, which identified potential interactors and compartment neighbours. The resulting data provides an unconventional insight into the vesicle trafficking and cargo sorting machinery in malaria parasites.
