| Description | In this project, we initially discovered the significant synergistic effects of vitamin C (Vit C) and Sulfamethoxazole (SMX) on various strains in both laboratory and clinical settings. To search for potential clues at the proteomic level for the synergistic property of Vit C+SMX observed in vitro, we quantitatively monitored changes in the proteome of cells treated with Vit C (1 mM), SMX (10 µg/ml), a mixture of both drugs at their respective concentrations, and a control group (no drug treatment). Mass spectrometry data were collected in Data Independent Acquisition (DIA) mode using a Q-Exactive HF X (Thermo Fisher Scientific, San Jose, CA) instrument for 36 samples (consisting of three biological replicates each of the above four groups of samples harvested after 12, 24, and 36 hours of treatment, respectively), quantifying a total of 37, 363 peptides and 2,685 proteins. Quantification of peptides and proteins was performed using MSstats software packages. After conducting enrichment analyses and functional annotation for proteins that were differentially expressed exclusively in the vc+smx group, we identified a consistent up-regulation of numerous proteins associated with copper homeostasis and redox stress. Coupled with the validation of a series of in vivo and ex vivo experiments, we elucidated the molecular mechanisms underlying the bactericidal effect of Vit C potentiation of SMX. |
