Title	Bioorthogonal Photocatalytic Proximity Labeling for the Profiling Golgi-associated Proteins in Living Cells
Description	Eukaryotic cells are characterized by a high degree of compartmentalization, wherein protein function is intricately tied to subcellular localization, as distinct compartments provide specialized microenvironments. The Golgi serves as a critical hub for the processing and sorting of proteins within intracellular trafficking pathways. The identification of Golgi-associated proteins in living cells is of great significance for understanding both intercellular and intracellular communication networks. However, this still poses a significant challenge because of the scarcity of targeted photosensitizers, which is attributed to the stacked membrane structure of the Golgi. Inspired by the mechanism of chondroitin sulfate accumulation in the Golgi of living cells, we prepared the Golgi-targeting nano-photosensitizer by combination of chondroitin sulfate and photosensitizers. The nano-photosensitizer enabled photocatalytic proximity labeling of the Golgi-associated proteins with high spatiotemporal precision under native conditions. After validation in living HeLa cells, which demonstrated that a substantial portion of Golgi-associated proteins could be identified with high specificity (77.8%), we applied the photocatalytic proximity labeling strategy to dynamic Golgi-associated proteins mapping of brain metastatic lung cancer cells. We found that Golgi-associated proteins played in signal transduction and extracellular matrix organization during cancer metastasis. Overall, this study not only significantly advanced the identification of the Golgi-associated proteins but also provided new insights into the potential functions of the Golgi.
