TitleSnx17 facilitates bunyavirus infection
DescriptionRift Valley Fever Virus (RVFV) is a negative-strand RNA bunyavirus with a tri-segmented genome and that can infect humans and animals due to its broad tropism. Through a loss-of-function genome-wide CRISPR Cas9 screen, we previously identified Lrp1 in RVFV entry facilitated by the viral glycoproteins. This screen also identified several additional host factors that are important for viral infection. Here, we describe our efforts to define a role for Sorting Nexin 17 (Snx17) in bunyavirus infection. Our findings reveal that the removal of Snx17 results in a substantial reduction of RVFV infection in Snx17 knock-out cells. Further analysis revealed that Snx17 removal leads to a reduction in the Lrp1 levels, but not complete loss of Lrp1 expression. We also show that Snx17 is essential for the entry and infection of several bunyaviruses that do not utilize Lrp1, suggesting a broader impact on viral entry. These results support a role where Snx17 can facilitate viral infection in an Lrp1-dependent and an Lrp1-independent manner. Our findings highlight how bunyaviruses use multiple host factors to gain cellular entry and identify additional pathways that can be used to dissect entry mechanisms and targeted for therapeutic intervention.
