| Description | Identifying regulators for tissue and organism regeneration is fundamental for regenerative biology and medicine. Most of the regenerative regulators studied by previous studies are derived from transcriptional changes, and few regulators have been screened for changes at the protein level. Here, we addressed this gap in knowledge by performing advanced mass spectrometry-based proteomics analyses of the model planarian Schmidtea mediterranea at 6 time points along regeneration. We established a spectral library covering approximately 10,000 proteins. Our proteomics data revealed upregulated ribosome biogenesis and ribosome, which was not observed in the transcriptome dataset but supported by Ribo-Seq analysis. Functional examination using RNA interference led to the discovery of 25 proteins essential for planarian regeneration. The protein abundance of ribosomal genes among them was significantly increased, while the transcription level showed a downward trend, and the translation level similarly showed an increasing trend. Troponin T was identified as a regulator of regeneration initiation that showed increased protein abundance before upregulation at the mRNA and translation level, suggesting a regulation of protein stability. Our study thus identifies ribosome-mediated and transcription-independent protein machinery essential for the initiation of planarian regeneration. |
