Description | Lignocellulosic biomass is an abundant and renewable resource that can be leveraged for the production of a diverse array of platform chemicals, including next generation biofuels such as isoprenol. One of the main obstacles for the efficient use of lignocellulosic biomass as a substrate are inhibitors formed during pretreatment that pose challenging for the development of efficient and cost-effective microbial processes. Acetate is a growth-inhibitory organic acid derived from O-acetylated hemicellulose and has high concentrations in many lignocellulosic biomass hydrolysates. In this work, we adapted Pseudomonas putida as an acetate-tolerant chassis for bioproduction with increasing concentrations of both the heterologous product (isioprenol) and acetate. Using this approach, we restored cell growth when acetate is provided as the sole carbon source. Our results show we possibly have rerouted the entry of acetate into a different step for entry into the TCA. We demonstrate the use of rational engineering and ALE to synergistically enable the development of robust production strains for the bioconversion processes with this abundant alternative substrate. |
