TitleComplexome profiling of subcellular fractions of Plasmodium falciparum gametocytes
DescriptionThe aim of the experiment was to get a better understanding of the complexome of Plasmodium falciparum gametocytes and its subcellular distribution. To do so we, whole parasite cells were isolated and subjected to nitrogen cavitation to free cellular content and homogenize the sample. To decrease sample complexity and obtain subcellular fractions, the homogenate was consequently separated through ultracentrifugation using a discontinuous sucrose gradient. The three resulting fractions were separately resolved by high-resolution clear native electrophoresis (hrCNE) followed by mass spectrometry and complexome profiling analysis using a novel analysis tool we developed, the Gaussian interaction profiler (GIP).
