| Title | The analysis of PRRSV nucleocapsid interactome |
| Description | The porcine reproductive and respiratory virus (PRRSV) is a significant swine disease, causing reproductive failure in sows and complex respiratory failure in sows and complex repiratory syndrome in pigs of all ages. PRRSV is an enveloped +ssRNA virus classified within the family Arteriviridae, which together with the family of coronaviridae constitutes the Nidovirales order. The viral genome encoded at least 20 proteins with nucleocapsid (N) protein being the most abundant viral protein in infected cells. Besides its primary role in packaging and protecting the viral RNA genome, the N protein has additional functions that can impact host cells. Investigating the N interactome can shed light on the molecular mechanisms of viral infection and pathogenesis, potentially leading to therapeutic advancements. Thus, we examined the PRRSV N interactome using EGFP-IP coupled to label-free quantitative mass spectrometry analysis and identified 301 N interactors. We found a significant enrichments of ribosome machineries, nucleolar proteins involved in ribosome biogenesis, splicing factors, RNA helicases, and DNA-binding proteins involved in chromatin remodeling and DNA damage response. Comparison of our data to coronavirus N interactomes revealed several interactions that are conserved among the Nidovirales family, such as tRNA ligase RTCB. Mining this dataset, our study expands the current landscape of PRRSV-host interactions and uncovers several N-interacting proteins that are new to PRRSV biology |
