| Title | LD4172 and LD4172NC treated MDA_MB_231 cell line global proteomics |
| Description | 1 million MDAMB231 cells were seeded in 6-well plates. On the following day, MBAMB231 cells were treated in triplicates with DMSO, LD4172 (100 nM) or LD4172-NC (100 nM) for 6 hours. Cells were washed with ice-cold PBS three times. The cell pellets were lysed, reduced, alkylated, and digested by EasyPep MS Sample Prep Kits (ThermoFisher: A45733). The same amount of peptide from each condition was labeled with tandem mass tag (TMT) reagent (ThermoFisher: 90113). The 10-plex TMT reagents were incubated with each peptide sample for 1 hour at room temperature. The labeled peptide samples were quenched by adding 50 uL of 5% hydroxylamine, 20% formic acid solution and mixed. Mixed samples were desalted and offline fractionated into 24 fractions as previously described. |
