TitleConditional SPI1 deletion exacerbates amyloid pathology through modulating microglial functions in a mouse model of Abeta amyloidosis
DescriptionRecent human genetic studies have highlighted microglia genes and its regulatory mechanisms involved in the pathogenesis of Alzheimer disease (AD). The transcription factor PU.1 (encoded by SPI1) is mainly expressed in microglia in the nervous system and has been reported as a genetic risk factor for AD. However, the role of microglial SPI1 on AD etiology is still poorly understood. Here, we first demonstrated that the loss of microglial Spi1 exacerbates amyloid pathologies in an AD mouse model (APP/PS1). Specifically, microglial Spi1 deficiency increases amyloid deposition, gliosis, dystrophic neurites, while decreasing plaque-associated microglia by modulating microglial phagocytic activity. Furthermore, we identified several potential biological networks, such as immune response_phagocytosis and Neurophysiological process, which are regulated by altered microglial Spi1 through the analyses of proteomics and transcriptomics data. Our findings broaden understanding of the role of microglial SPI1 in amyloid pathology and provide a rich dataset for future analyses of SPI1 in AD.
