Dataset Summary
TitleXenopus laevis sperm extract IEX and SEC Fractionation
DescriptionSperm were flushed from freshly dissected Xenopus laevis (J strain) testis using Marks Modified Ringers solution, and collected by centrifugation. Purified sperm were lysed in buffer containing 1% n-Dodecy-beta-D-Maltoside and the clarified extract was fractionated on either a mixed bed ion exchange column or an SEC. Column fractions were reduced, alkylated, and digested with trypsin for mass spectrometry as follows. IEX samples were run on an Orbitrap Fusion using a 75 minute topspeed CID method, while SEC samples were run on an Orbitrap Fusion Lumos using a 75 minute topspeed HCD method. Data analysis was using the MSBlender pipeline, and an Xenla database collapsed at the eukaryote level using eggNOG mapper.
