TitleIntegrative biochemical and proteomics analysis reveals the toxic mechanisms of shale gas flowback water on algae Chlorella pyrenoidosa
DescriptionThe toxic mechanisms of flowback water (FBW) need to be concerned due to their high production amount but is still not identified due to their excessive complex constituents. In this study, the toxic mechanism of FBW were clarified using ibiochemical and proteomics analysis. Firstly the toxicity of 11 different FBW on Vibrio fischeri luminance was found to be mainly affected by their organic contents. Then, three typical FBW were selected to further evaluate their toxicity on the growth of green algae Chlorella pyrenoidosa (C. pyrenoidosa), and the toxicity of the respective activated carbon treated FBW (AC-FBW) with partial organic compounds removed and equivalent salt control (SC) were also tested to discern the effect of organics, inorganics, and salt of FBW. The growth inhibition toxicity of the three FBW is negatively related with the organic content and positively with metal content. And, all the AC-FBW showed the strongest inhibition toxicity, followed by SC and original FBW. Extracellular LDH activity test, SEM morphology observation, and oxidative stress indicators analysis also demonstrated that AC-FBW showed strongest toxicity on cell membrane and oxidative damage. All the biochemical tests indicated that the inorganics in the FBW play primary role in their toxicity on algae growth, which is further verified by the toxicity test of stimulated FBW. Then, proteomics revealed that FBW and AC-treated exposure significantly influenced different multiple proteins, mainly involved in the metabolism. Meanwhile, the proteins associated with metal and salt stress were found to be up-regulated more significantly in cells exposed to AC-treated FBW.
