We used a proteomics-guided approach to identify and annotate the proteins enriched in the vigorous camptothecin metabolism period. For transition library construction, shotgun proteomics analyses were performed using Evosep One UHPLC system coupled with a Q ExactiveTM HF-X mass spectrometer operating in the data-dependent acquisition and data-independent acquisitionmode. The resulting spectra from each fraction were searched separately against the genome database of C. acuminata by the search engines: Proteome Discoverer software (version 2.2). The search parameters were set as follows: mass tolerance for precursor ion was 10 ppm and mass tolerance for product ion was 0.02 Da. The result of search and identification by PD 2.2 software was imported into Spectronaut software (version 14.0) to generate a library.
