TitleSpatially region resolved proteomic map reveals extracellular matrix regulates epidermal growth and function as epidermal stem cell niche
DescriptionSpatial proteins expression is critical to identify exactly protein localization and function in tissues. By combining decellularization, laser-capture microdissection, and mass spectrometry, six different partitioned proteins among skin layers of stratum corneum (SC), granular-spinous (GS), basal layer (BL), BM, superficial dermis (SD), and deep dermis (DD) were isolated, enriched and for the first time, a stratified and developmental lineage proteome map of human skin has been revealed spatially. Especially, we found TGFβ induced (TGFBI), an ECM glycoprotein, in the BM that enhances the proliferation of EpSCs and promotes the process of wound healing. Most interestingly, TGFBI shows the potential for treating secondary syphilis patients, which may benefit clinical practice.
