Hypothyroidism model rats were established with PTU (10 mg•kg-1•d-1) and treated with Atractylodis Macrocephalae Rhizome and its fractions (volatile oil fraction, lactones fraction, atractyloside fraction, oligosaccharide fraction, and crude polysaccharide fraction) for 20 d. The experiment (having been) finished, liver samples were gained. Protein concentrations measured by the BCA kit and the tryptic peptides obtained from the different groups were labeled with iTRAQ reagents. The peptides were eluted into 10 fractions by High-pH Reverse-phase Liquid Chromatography Fractionation and analyzed by LC-ESI-MS/MS Analysis based on Triple TOF 5600. Results indicated that these DEPs contained Pygl, Pklr, Pc, Ehhadh, Acox1, Fasn, Acly, Acsl1, Acsl5, Pgm1, Suclg1, Gpt, Idh1, Fh, and Adh1, which could represent target proteins of Atractylodis Macrocephalae Rhizome and its fractions for hypothyroidism. Atractylodis Macrocephalae Rhizome and its fractions played roles in regulating the substance and energy metabolism for the treatment of hypothyroidism.
